In my opinion, no. They are basically the same thing, except Ultimate adds things like dashing, hanging off ledges and of course, characters. Also, the online play adds a ridiculous amount of depth, which is absent in Super. The koma (the characters you fight with and support characters) are unlocked a lot easier for English-speaking players. In Super, you have to match a character with a silhoutte and a quote, which can be easy to ridiculously difficult. Ultimate has an easy to handle type of evolution tree thing in which you use points from fighting to get more koma.

Yes, but it doesn't offer many options. I think you can only choose between Naruto and Luffy and one level, I don't remember which. In Ultimate, you have slightly more possible characters, but they're picked at random.

I would say you'd probably still enjoy it, since it really is a good game. Of course, liking the manga that are featured helps, and I know I definitely wouldn't enjoy it as much if it didn't have the stuff I like. Plus, it introduced me to some new ones like D. Gray-Man and Gintama, both of which I like now. The biggest thing for most (English) gamers is of course the fact that it's in Japanese, but if you can get past that you'll enjoy it.
