I don't use Twitter -- and I still regard it as a shithole -- but I wasted the better part of an hour reading Brooks, Martin, and Shandling's tweets.
I found about it through a recent AV Club profile of Brooks, which pointed out that he's on Twitter. Brooks treats twitter like he's playing to a room, like he's on the Tonight Show with Johnny. Shandling (and Martin to a lesser extent) is the same way with his twitter account, more often than not he has this compulsion to be "on".
I checked out a couple of the comedians that those guys follow (e.g. Sarah Silverman, Jean Ralphio from PNR, Kevin Pollack), and while those guys try, they can't match the quality of Brooks, Shandling, or Martin. Brooks especially is hilarious.
