Wireless router needing wireless adapter

Here's the scoop: My sister and her husband recently moved and want to set up a wireless network connection. They own this (or something just like it; I know it's Netgear):

My question: If their modem is on the third floor and one of their desktops is in the basement (1st floor), will a USB network adapter like this work?

Or would an actual internal wireless card be a better option? Like this:

My only problem with the internal card (because I'd be installing it) is that I can't tell if the empty slots are PCI or PCI Express. I opened system settings and only saw listings for PCI Express slots, but that was only for three and there are five on this PC.

Damn. I knew I should have told him to wait. My brother-in-law wanted something as soon aspossible, so I just told him to go with the wireless one in case he bought a new computer or wanted to use it with a laptop that didn't have wireless. Thanks anyway, though. Now I know for future reference.
