Should I be concerned if my DS cartridge sounds loose? It makes a rattling sound when I shake it. If I apply a little pressure to the face and back of the cart the noise stops.

All my DS games vary in rattle from nothing (Bleach) to quite rattly (Custom Robo Arena).

I'm thinking it mostly has to do with the thickness of the chip on the board, considering the fact that there's no actual screws on a DS cart to keep it extra tight.

Here's an excellent little Youtube video I found showing the inner chip, so we don't have to bust open our own unwanted cart (lol @ him using Gunpey as his guinea pig):

Notice the slotted holes on the sides of the chip? they're giving it the ability to rattle, but the two fitted holes near the middle top keep it completely secure.

BE WARY OF RATTLING CARTRIDGES! I got a rattling copy of Phoenix Wright: Justice for All from Goozex, and it turns out that the contacts can shake loose, even when the cart is in the DS. This means that if you move because your arm is tired, your game might freeze up (which happens ALL THE TIME with my copy of PW:JFA) What's worse is there is a chance that it can short your system, which is muy no bueno.

Question is, does anyone know how to tighten DS carts?

Those notches on the side keep it in place too, if you take a close look at the inner casing, the correspond with pegs in the mold. Any rattling comes from the case pulling loose.

If you can get the case off without busting it such that you can put it back on, a drop or two of glue should hold it down secure for the rest of eternity.
