I used the default Sheploo and femShep, though that was just out of laziness.
If you have a class where SMG is your best long-range weapon, like an Adept, getting the Locust SMG at the end of Stolen Memory is a must. It's the best SMG in the game, so it's well worth the time spent on it to get it, even if your class doesn't rely on it for its main long-range weapon. Plus, IMO, I think the end part of the game is very good combat, so that is another reason to do it.
You can't do LoTSB until you've gone to Illium and talked with Liara and found the Shadow Broker agent in her midst, which prompts her to go after the Shadow Broker and find them. That'll be the lead-in to LoTSB, so you'll end up picking up some crewmates and doing some missions before you get to that point. It's useful for the additional power you get available to you from Liara as well as the ability to re-spec crewmates' abilites after completing LoTSB.
Overlord can be done at any time and it doesn't give you any in-game benefits. Supposedly what you do at the endgame of Overlord will be one of those decision points that shapes ME3's game.
Arrival should be done last at the end of the game after doing everything, since that's how it's written from a story perspective. It's timing in-game of when it can be done is around the middle of the normal gameflow timeframe, so you can do it around the middle of the game or later.
Picking up the Firepower pack is also a good idea, since it gives you the best pistol in the game, the best assault rifle in the game and also the best shotgun in the game. Well worth the 160 MSP spent on it.
Oh, I know of the magic of the Locust, I'm a Vanguard and it's my weapon of choice
Wow, I'm really surprised that Overlord is a big deal, if I'd known that it supposedly shapes ME3 then I would've gotten in a long time ago. I just finished Arrival for my original playthrough and think it fits well into the end of the game (after the suicide mission) so I'll keep that there and I'm downloading the Firepower pack now. Thanks a lot, Shrike!
Also, you should totally customize your Shep! It's a lot of fun! Be careful if you ever wanna go back and tweak him/her, though. It's really easy to mess them up. I've tried editing my Sheps a couple of times and I almost always mess up really badly on the first try. Luckily I always have a copy of my save to put back onto my 360 and restart from.
