In all honesty, I've beaten the game four times since finals week and I'm still not done with it. I don't see how anyone could go into Mass Effect 2 without experiencing the first game (but they say it happens, so who knows..).
At the very least play the game one time through. Do as many sidequests as you can get your hands on (look up a guide if you have to) and get very familiar with the game. Checking out the codex is also a very good source of information - it's not just some gimmicky thing they through in the game. Even powering through ME1 won't give you the full idea of what's to come in ME2. The books (specifically,
Mass Effect: Ascension) provide somewhat of a smooth transition between the two games, and also give more information about a couple characters that will appear in ME2.

They've already stated that you can customize your armor, ammo upgrades are now part of the bumper wheel so you can change ammo types on the fly, you receive weapon upgrades from completed missions and from sidequests where you bring back parts/research to the Normandy, and that you change out your weapons/equipment before the start of any/each mission. What else is there to know?

these videos sooner (videos called "Savage" and "Assassin" for Grunt and Thane, respectively). I have no idea what this new "Fight For The Lost" thing is, but being able to see more footage of those two is more than enough for me.
